The binding of carbon monoxide by human hemoglobin. Proof of validity of the spectrophotometric method and direct determination of the equilibrium.

چکیده
Stoichiometric titrations of human hemoglobin with carbon monoxide show that the interaction of hemoglobin with ligand is accompanied by proportional changes in both the Soret and visible absorption bands. This linearity has been observed in neutral solutions, both in the presence and absence of 2 M NaCl, and in alkaline solutions at pH 9.2. It is concluded that the liganded intermediates taking part in the equilibrium do not differ significantly in spectra from the equivalent hypothetical mixtures of reduced and fully liganded hemoglobin. The CO equilibrium determined in dilute hemoglobin solutions (4.5 X 10dl M to 2.34 X lo-’ M) is characterized by a relatively high Hill constant (2.3) and a free ligand concentration of 4.3 to 5.6 x lo-* M at 50% saturation. The results are of special interest in light of the sedimentation studies and kinetic experiments which indicate that a strongly ligand-linked equilibrium between dimeric and monomeric (or single chain) hemoglobin predominates in this concentration range.

A modification of the spectrophotometric determination of carbon monoxide in blood enabling direct determination of per cent hemoglobin saturation.
BECAUSE OF VARIATION in hemoglobin concentration in normal bloods with age and sex (1), the per cent carbon monoxide saturation of hemoglobin represents a more accurate toxicity index than the volume per cent of carbon monoxide. In 2 of the most common methods of carbon monoxide determination in blood, the gasometric method of Van Slyke (2, 3, 4) and the spectrophotometric method of Harper (5),...
متن کاملDetermination of mercury (II) by spectrophotometric method based on self assembly of gold nanoparticles
Due to the binding of the –COO- group with the mercury (II), gold nanoparticlesfunctionalized with glutathione can self-assemble to form a supermolecular network in the HAc-NaAc buffer solution. The aggregation of functionalized gold nanoparticles and mercury (II)caused the shift of maximum absorption peak of the UV-Vis spectrum from 520 nm to 609 nm.Based on this principle, a simple spectropho...

Little use has thus far been made of the near infra-red spectrum for the identification and determination of hemoglobin and its derivatives BJ means of infra-red photography Eggert (1) established the transparency of carbon monoxide-saturated blood and used this property for the qualitative detection of carbon monoxide poisoning.. متن کاملExtraction - Spectrophotometric Determination of Palladium with Tetrabutyl Ammonium Iodide
A sensitive and selective extraction - spectrophotometric method for the determination of trace amounts of palladium is reported. The method is based on quantitative extraction of brownish tetrabutylammoium tetraiodopalladium (II) ion pair into a small valume of chloroform at pH 1.3-12.0, followed by spectrophotometric measurements. The Beer's law is obeyed over the range 0.02-1.09 <em...
متن کاملPartial Least Square and Parallel Factor Analysis Methods Applied for Spectrophotometric Determination of Cefixime in Pharmaceutical Formulations and Biological Fluid
In this study, the direct determination of cefixime as an anti-bacterial agent, in pharmaceutical formulations, urine and human blood plasma was conducted based on spectrophotometric measurements using parallel factor analysis (PARAFAC) and partial least squares (PLS). The calibration set was composed of fourteen solutions in the range of 0.50- 9.00 µg mL-1. PLS models were calculated at each p...
